X-Glu ist Substrat der ?-Glucosidase und entspricht dem X-Gal als Substrat der ?-Galactosidase, die durch Spaltung des Substrates 5-Brom-4-chlor-3-indoxyl (X) freisetzt. Das enzymatisch abgespaltene Indoxyl wird zum unlöslichen Indigo oxidiert.
