Tth DNA Polymerase ist ein rekombinantes Enzym zur hocheffizienten Amplifikation. Es ist eine 5' - 3' DNA Polymerase ohne 3' - 5' Exonucleaseaktivität.

Genaxxon BioScience Tth DNA Polymerase ist die rekombinate Form eines Enzymes das aus dem thermophilen Eubakterium Thermus Termophilus (HB-8) stammt. Das Enzym ist eine hochprozessive 5' - 3' DNA Polymerase die keine 3' - 5' Exonucleaseaktivität zeigt. In Gegenwart von MgCl2 katalysiert Tth DNA Polymerase die Polymerisation von Nukleotiden in Dublex-DNA in 5' - 3' Richtung, doch zeigt die Tth-Polymerase eine sehr hohe intrinsische Reverse Transkriptase Aktivität (RT) in Gegenwart von Mn-Inonen. Die RT Aktivität ist nicht mit einer RNase H Aktivität verknüpft. Durch Verwendung von Tth-Polymerase an Stelle einer normalen RT minimiert Probleme die im Zusammenhang mit Sekundärstrukturen auftreten können, da die reverse Transkription bei höheren Temperaturen durchgeführt werden kann. Tth ist auch bei Temperaturen über 95°C über längere Zeit stabil.
