Recombinant mouse TPO produced in E.Coli is a single, non-glycosylated soluble polypeptide of 174 amino acids and a molecular mass of 18704 Dalton. Thrombopoietin is a glycoprotein hormone produced mainly by the liver and the kidneyt hat regulates the production of platelets by the bone marrow. It stimulates the production and differentiation of megakaryocytes, the bone marrow cells that fragment into large numbers of platelets. ED50 range = 0.05-0.015ng/mL corresponding to a specific activity of 20-66MU/mg, determined by the dose dependent proliferation of Mo7e cells. The optimal concentration for each specific application should be determined by an initial dose-response assay.
